Due to development of duck production in Russia it has become necessary to control duck diseases such as duck virus hepatitis (DVH) which is the most economically important disease.
DVH is a highly contagious disease characterized by liver lesions and high lethality of ducklings. DHAV-1 is the most widely distributed and important serotype for disease diagnostics and prevention.
The reference laboratory for viral diseases of the subordinate to the Rosselkhoznadzor FGBI “ARRIAH” developed the first test-system based on real time PCR (real time polymerase chain reaction) combined with real time RT-PCR (Reverse transcription polymerase chain reaction) for detection of DHAV-1 genome. New primers and a probe were designed for DHAV genome detection with real time one-stage RT-PCR. Parameters (reagent concentration in reaction mixture and PCR temperature cycle parameters) for reaction using home-produced enzymes in rapid two-stage real time PCR were optimized. Analytical sensitivity of the optimized method was close to the previously published RT-PCR method and was 100 ELD50/cm3 (LD50/cm3).
The developed system was successfully tested using home strains of duck virus hepatitis agent.
