Patent of the Russian Federation for invention was obtained by stuff-members of the Reference Laboratory for Rabies and BSE, Laboratory for Foot-and-Mouth Disease Prevention, Reference Laboratory for ASF and Laboratory for Molecular and Genetic Research of the FGBI "ARRIAH" (Vladimir) subordinated to the Rosselkhoznadzor.

The document certifying the sole right and authorship of specialists of the Federal Center for Animal Health (FGBI “ARRIAH”) has been issued for:

“Method for differentiation of genetically modified rabies virus ARRIAH-G333 strain genome and rabies virus vaccine RV-97 strain genome with real-time polymerase chain reaction and high-resolution peak analysis”.

Rabies is a fatal disease of animals and humans that causes great economic losses due to the direct economic damages, as well as the costs of the measures to be taken for the disease prevention. The causative agent of the disease is the rabies virus, member of Rhabdoviridae family, Lyssavirus genus.

Vaccines of various types including live ones are used for rabies prevention. The strain avirulence for the target animal/human is a crucial for its use for live vaccine production. The strain carrying a mutation contributing to reduced virulence in its genome has been developed using genetic technologies.

Development of genetic constructs for specific prevention makes important methods for their detection. The methods must be highly specific and sensitive.

High resolution melting curve analysis of DNA fragment extracted by polymerase chain reaction (HMR-PCR) enables differentiation of DNA-targets with multi-nucleotide substitutions.

The authors proposed a method based on melting curve analysis capable of detecting the genome of a genetically engineered rabies virus with an attenuating mutation in G333 glycoprotein gene and differentiating it from wild-type variants in any samples of biological materials of animal origin.
